K\u1ef9 thu\u1eadt \u201csimple and quantitative PCR\u201d, ph\u00e2n t\u00edch Southern, th\u1ef1c hi\u1ec7n \u201cgenome walking\u201d, v\u00e0 s\u1eed d\u1ee5ng th\u01b0 vi\u1ec7n BAC (bacterial artificial chromosome library) \u0111\u01b0\u1ee3c \u00e1p d\u1ee5ng trong thanh l\u1ecdc v\u00e0 gi\u1ea3i tr\u00ecnh t\u1ef1 m\u1ed9t c\u00e1ch r\u00f5 r\u00e0ng. K\u1ebft qu\u1ea3 ch\u1ee9ng minh r\u1eb1ng hai v\u00f9ng T-DNA kh\u00e1c nhau (IbT-DNA1v\u00e0 IbT-DNA2) \u0111\u1ec1u th\u1ec3 hi\u1ec7n trong genome c\u1ee7a gi\u1ed1ng khoai lang tr\u1ed3ng tr\u1ecdt (Ipomoea batatas [L.] Lam.). Nh\u1eefng gen ngo\u1ea1i lai (foreign genes) n\u00e0y th\u1ec3 hi\u1ec7n \u1edf m\u1ee9c \u0111\u1ed9 c\u00f3 th\u1ec3 ph\u00e1t hi\u1ec7n \u0111\u01b0\u1ee3c (detectable) trong c\u00e1c m\u00f4 kh\u00e1c nhau c\u1ee7a khoai lang. IbT-DNA1 \u0111\u01b0\u1ee3c t\u00ecm th\u1ea5y c\u00f3 t\u1ea5t c\u1ea3 b\u1ed1n khung \u0111\u1ecdc m\u00e3 gi\u1ed1ng nhau (ORFs: open reading frames) \u0111\u1ed1i v\u1edbi c\u00e1c gen tryptophan-2-monooxygenase (iaaM), indole-3-acetamide hydrolase (iaaH), C-protein (C-prot), v\u00e0 agrocinopine synthase (Acs) c\u1ee7a vi khu\u1ea9n Agrobacterium spp.<\/p>\n<p>IbT-DNA1 \u0111\u01b0\u1ee3c t\u00ecm th\u1ea5y trong t\u1ea5t c\u1ea3 291 gi\u1ed1ng khoai lang tr\u1ed3ng tr\u1ecdt kh\u1ea3o nghi\u1ec7m, nh\u01b0ng kh\u00f4ng g\u1ea7n v\u1edbi c\u00e1c lo\u00e0i hoang d\u1ea1i. IbT-DNA2 c\u00f3 ch\u1ee9a \u00edt nh\u1ea5t n\u0103m ORFs v\u1edbi s\u1ef1 \u0111\u1ed3ng d\u1ea1ng c\u00f3 \u00fd ngh\u0129a (homology) \u0111\u1ed1i v\u1edbi ORF14, ORF17n, locus qui \u0111\u1ecbnh t\u1ea1o r\u1ec5 (Rol)B\/RolC, ORF13, v\u00e0 ORF18\/ORF17n c\u1ee7a vi khu\u1ea9n A. rhizogenes. IbT-DNA2 \u0111\u01b0\u1ee3c t\u00ecm th\u1ea5y \u1edf 45 gi\u1ed1ng trong s\u1ed1 217 m\u1eabu gi\u1ed1ng khoai lang kh\u1ea3o nghi\u1ec7m v\u00e0 lo\u00e0i khoai lang hoang d\u1ea1i.
